Could you give me a link to the headset you are using? Did it have USB and you use the line-in from the Mac or is it with / like a external Soundcard?

THXs for the information. I think you had the same issue like me. I found a workaround for that:
The problem is that an external soundcard make problems. I did not know why, but if you use it with the OS or Skype it worked fine with USB but if you use it with x-plane 10.20 (32 bit) you had the issue as described.
If you make a test with the internal mic it will works. But this is not what we like to do... we had sound on the speakers and it did not work well with the internal mic.
The next problem is the iMac. It has a line-in and no mic-in. That means that you need a source for the line-in that has power - like a iPod or CD-player. When you try to use the plantronics direct (without USB) I think it will not work. Not in the OS (Skype) and also not in x-plane.
The workaround is following:
option 1: you need a Headset with power / battery inside or power over USB
option 2: you use your headset with a mic pre amp and than use the line-in form the iMac
I did it with the second option. But if you did not want to have the airplane sound on the headset you had to use external speakers with a external usb sound card. This is the way I did it. System audio on the external sound card / speaker. And the headset plugged in the audio out and line-in (with pre amp). Than you had to set in the plugin config the internal sound card as sound-source.

First of all let me thanks you for the infos.
Now I understand what's going on, but still have no chance to avoid the issue with my actual hardware.
Can't even try to connect my headset directly to the imac without usb because my Plantronics is an old one and has no jacks (just usb).
Anyway it seems laminar xplane 10.20 related so maybe we can try to post them.
As matter of fact I'm using same hardware on 10.11 and it works flawless and definitely I don't want same issue on future xplane 10 versions.
I have and external usb sound-card (to connect the external speakers) but prefer to have the audio of the plane on my headset so I am not disturbing my children sleeping.
(Another try will be disconnect this external usb sound card to avoid any kind of conflicts).
Can you suggest me an headphone with amplificated mic?
This way I can follow your workaround and connect it directly to the imac by jacks.
Thanks in advance

I had no experience with amplificated mic. I use a pre amp like this:
http://www.amazon.com/Behringer-MIC800- ... amplifiers
The Behringer is bigger than needed - but it works. And you can use each passive headset.
If you had to by a new headset - by one with USB-Power or Battery-Power. Both must work with a Mac.
Maybe the Sennheiser U 320 also work with Mac. But as I said - I had no experience:

Spending a couple of hours investigating the issue and have to admit have no clues.
Facts:
1 - no one mic is working, tried also with internal one. Last chance is to try with imac line-in but it sounds a bit strange that 10.20 forces input sound only to the line-in.
2 - it's not OS related. It's still present using the previous OS X.
3 - it's 10.20 specific and not present on any other version, xplane 9 included.
Dunno if it's can be related to the PE plugin neither I know if will be solved by the new mac 64bit library Keith is working on.
Confused.

I'm reporting the only 10.20 audio related modification started with 10.20 release 3, to avoid Gizmo ans SASL conflict.
From x-plane.com I paste
X-Plane 10.20 Beta 3
We still have a number of open bugs; please do _not_ re-report bugs that you have already filed against beta 2 unless you see them marked as fixed in this list! We will be getting to all of the filed bugs shortly, but this build gets out a few changes that we needed to test separately.
OpenAL sound is now initialized before any plugins are loaded. This changes which OpenAL implementation will be loaded for globally installed plugins. Please see the OpenAL technote for details. This fix should conflicts between Gizmo-enhanced and SASL-enhanced airplanes.
Link to the OpenAL technote

Maybe here is the solution and this is why xplane 10.20 pretends the imac line-in audio.
Anyway I'm still convinced that there is a way to let xplane uses other OpenAL devices.

Sounds like different issues. For me this workaround works and also I am / was able to use the internal mic of my 27" iMac.
Similar is that I can't use the USB external Soundcard and that it starts with 10.20 for me. Before the external Soundcard works well with x-plane 10.10

Later will try again disconnecting all external sound cards (I have Plantronics USB headset and an external nuForce DAC piloting my audio system) and transmit on Pilot Edge with internal mic.
The problem is I don't want to bother ATC but have no other choice.
After all if I can't transmit on voice they can't hear me, so I'll not bother anyone.
Pity we haven't a test frequency to setup our systems.
To be sure that my line in mic works I should transmit something from mi iPod (with preamplificated line).
Maybe will do that for 2 or 3 seconds on a non towered airport and that will hear the test.
In this case will buy and external ampli to pilot a mic (which I have but need a phantom connection) and will connect a passive headset too.
will go on posting the results.
thanks again
Angelo

Yes Goose,
You were absolutely right.
It seems that 10.20 accept only the default/integrated sound card; maybe Keith on the PE plugin can let the user decide about which audio device use but at the moment it's not possible.
So your workaround works to me too, and maybe I can add another one.
The imac headphone jack-in let you use passive earphone with microphone (iPhone earphone for example).
It works great and sound is not that bad.
Other chance is internal microphone (it works in my imac too).
Anyway I'm not leaving the 10.11 till Keith will release the 64bit version of the PE client. (Hope soon)
